The Virgin boss paid tribute on Tuesday to Joan Branson, née Templeman, whom he had been with for 50 years.
She was born in Glasgow and has two children with Sir Richard.
The pair got married at Necker Island, a 30-hectare island in the British Virgin Islands, which is owned by the businessman.
Sir Richard Branson said on Instagram: “Heartbroken to share that Joan, my wife and partner for 50 years, has passed away.
“She was the most wonderful mum and grandmum our kids and grandkids could have ever wished for.
“She was my best friend, my rock, my guiding light, my world.
“Love you forever, Joan x.”
