At Around 6.50pm, the man entered the Possil Road branch of Totesport and threatened the staff. They handed over a small quantity of cash, which the suspect took before fleeing.
No-one was injured, but staff were left extremely shaken from the experience.
The police were alerted, and firearms officers arrived out an immediate search of the area, but there was no trace of the suspect.
Detectives have been speaking to neighbouring businesses and properties to find out if anyone saw anything or if they have any information that could help them identify the man responsible.
He is described as a white man who was wearing a black jacket, a black hat and a scarf.
Detective Constable Gerry Hawkes from Strathclyde Police said: "Fortunately no-one was injured on this occasion, although members of staff are very upset and distressed by what has happened."
"I would appeal to anyone who may have been in or around the bookmakers at the time of the incident to get in touch with us. We need to trace this man and any information could help us in tracking him down."
